Helicobacter pylori lipopolysaccharide structural domains and their recognition by immune proteins revealed with carbohydrate microarrays

The structural diversity of the lipopolysaccharides (LPSs) from Helicobacter pylori poses a challenge to establish accurate and strain-specific structure-function relationships in interactions with host. Here, LPS domains five clinical isolates were obtained compared reference strain 26695. This was achieved combining information analysis (GC-MS ESI-MSn) binding data after interrogation LPS-derived carbohydrate microarray sequence-specific proteins. All LPSs expressed Lewisx/y N-acetyllactosamine determinants. Ribans also detected all isolates, allowing their distinction 26695 LPS. There evidence for 1,3-d-galactans blood group H-type 2 sequences two latter not yet described H. Furthermore, analyses showed strain-associated recognition by immune lectins DC-SIGN galectin-3 revealed distinctive patterns IgG antibodies serum pylori-infected patients.
